B細胞活化因子（BAFF）是TNF配體超家族成員，由巨噬細胞，樹突狀細胞和T淋巴細胞產(chǎn)生。BAFF促進B細胞的存活，對B細胞成熟至關(guān)重要。BAFF與三個TNF受體超家族成員結(jié)合：B細胞成熟抗原（BCMA / TNFRSF17），跨膜激活劑和鈣調(diào)節(jié)劑以及親環(huán)蛋白配體相互作用子（TACI / TNFRSF13B）和BAFF受體（BAFF R / BR3 / TNFRSF 13C）。這些受體是缺乏信號肽的III型跨膜蛋白。雖然TACI和BCMA結(jié)合BAFF和另一個TNF超家族配體APRIL（一種增殖誘導配體），BAFF R選擇性結(jié)合BAFF。BAFF R細胞外結(jié)構(gòu)域缺乏TNF受體典型的富含半胱氨酸的結(jié)構(gòu)域（CRD），并且僅包含具有四個αysteine殘基的部分CRD。人類和小鼠BAFF R共享56%的身份。BAFF R在脾臟、淋巴結(jié)和靜息B細胞中高表達。它也在活化的B細胞，靜息CD4 + T細胞，胸腺和外周血白細胞中以較低的水平表達。
B-cell activating factor (BAFF) is a TNF ligand superfamily member and is produced by macrophages, dendritic cells, and T lymphocytes. BAFF promotes the survival of B cells and is essential for B cell maturation. BAFF binds to three TNF receptor superfamily members: B-cell maturation antigen (BCMA/TNFRSF17), transmembrane activator and calcium-modulator and cyclophilin ligand interactor (TACI/TNFRSF13B) and BAFF receptor (BAFF R/BR3/TNFRSF 13C). These receptors are type III transmembrane proteins that lack a signal peptide. Whereas TACI and BCMA bind BAFF and another TNF superfamily ligand, APRIL(a proliferation-inducing ligand), BAFF R selectively binds BAFF. The BAFF R extracellular domain lacks the TNF receptor canonical cysteine-rich domain (CRD) and contains only a partial CRD with four αysteine residues. Human and mouse BAFF R share 56% identity. BAFF R is highly expressed in spleen, lymph node and resting B cells. It is also expressed at lower levels in activated B cell, in resting CD4+ T cells, in thymus and peripheral blood leukocytes.
