ScienCell's GeneQuery™ Human Myeloid Lineage Markers qPCR Array Kit (GQH-MLM) is designed to facilitate gene expression profiling of 40 marker genes involved in myelopoiesis. In the tightly regulated lineage of common myeloid progenitors (CMPs), the CMPs can first differentiate into granulocytes, monocytes, mast cells and macrophages. The granulocytes can further differentiate into mature eosinophils, basophils, and neutrophils. Brief examples of how included genes may be grouped according to their functions are shown below:     

·         CMP markers: CD34, CD38, KIT, FLT3

·         Granulocyte-monocyte precursor markers: CD34, CD38, CD48, IL3RA, FLT3

·         Monocyte markers: ITGAM, CD14, PECAM1, SELL, CSF1R, CCR2, FCGR3A, FCGR3B, CXCR3

·         Macrophage markers: ITGAM, ITGAX, CD14, CD68, CD80, CD163

·         Polarized macrophage markers: CD68, CD86, TLR2, TLR4, CIITA, CD163, CD200R1, MRC1, CD81, VCAM1, CLEC7A, VEGFA

·         Mast cell markers: FCER2, KIT, ENPP3, FCER1A

·         Neutrophil markers: ANPEP, FUT4, CD33

GeneQuery™ qPCR array kits are qPCR ready in a 96-well plate format, with each well containing one primer set that can specifically recognize and efficiently amplify a target gene's cDNA. The carefully designed primers ensure that: (i) the optimal annealing temperature in qPCR analysis is 65°C (with 2 mM Mg2+, and no DMSO); (ii) the primer set recognizes all known transcript variants of target gene, unless otherwise indicated; and (iii) only one gene is amplified. Each primer set has been validated by qPCR with melt curve analysis, and gel electrophoresis.
